What are the side effects of Tukysa (tucatinib)?

The main side effects of Tukysa (tucatinib) are: nausea, vomiting, mouth ulcers/pain, loss of appetite, weight loss, fatigue, or muscle/joint pain may occur.

Diarrhea is a common side effect and can sometimes be serious. Continuous vomiting/diarrhea may lead to a large loss of body water (dehydration). If you notice any symptoms of dehydration, such as abnormally decreased urination, dry mouth/thirst, fast heartbeat, or dizziness/lightheadedness, please contact your doctor immediately. Your doctor may prescribe antidiarrheal medicine, which should be taken during treatment.

People who use Tukysa (tucatinib) may have serious side effects. However, your doctor prescribes this medicine because he/she believes that the benefits to you outweigh the risks of side effects. Careful monitoring by a doctor may reduce your risk.

Tukysa (tucatinib) may cause you to have a skin problem called hand-foot syndrome. To avoid this, please protect your hands and feet from heat or pressure. Avoid activities such as using hot water, washing the bathtub, jogging, walking long distances, or using garden or household tools (such as screwdrivers). Symptoms may include pain, swelling, redness, blisters or numbness in the hands/foot. Your doctor may prescribe medications (such as lip balm) to help relieve symptoms. If symptoms affect your daily activities, seek medical help immediately.

Tukysa (tucatinib) may rarely cause serious (possibly fatal) liver disease. Your doctor should check liver function tests before starting this medication and every 3 weeks or as needed during treatment. If you have any symptoms of liver damage, please seek medical help immediately, such as: nausea/vomiting, stomach/abdominal pain, yellow eyes/skin, dark urine.

Very severe allergic reactions to Tukysa (tucatinib) are rare. However, if you notice any symptoms of a severe allergic reaction, including skin rash, itching/swelling (especially face/tongue/throat), severe dizziness, and difficulty breathing, please seek medical help immediately.

How to use Tukysa (tucatinib)?

Follow your doctor’s instructions to take this medicine by mouth, with or without food, usually twice a day (about 12 hours apart). Should be swallowed whole, do not chew, crush or split the tablet. If the tablet is cracked or damaged, do not take it.

If you miss a dose or vomit after taking Tukysa (tucatinib), please do not take another dose, just take the next dose at the regular time.

Use this medication regularly to get the most benefit. To help memory, take it at the same time every day.

The dosage depends on your medical condition and response to treatment. Do not increase the dose, and do not use this medication more frequently or for a longer period of time, otherwise your condition will not get better soon, but the risk of serious side effects will increase.

Because Tukysa (tucatinib) can be absorbed through the skin and lungs and may harm the unborn baby, pregnant women or women who may become pregnant should not use the drug or inhale the dust in the tablet.

This medication may slow the clearance of other medications from your body, which may affect the way they work. Examples of affected drugs include avanafil, certain benzodiazepines (such as midazolam, triazolam), ergot medicines (such as ergotamine, dihydroergotamine), and certain “statins” Drugs (such as lovastatin, simvastatin), ubrogepant, etc.

Do not share this medicine with others.

Laboratory and/or medical tests (for example, complete blood count, kidney/liver function) should be performed before and during the start of this medication.

Missed dose

If you miss a dose, skip the missed dose. Take the next dose at the regular time. Do not double the dose to catch up.

Storage 

Store tightly closed in the original container at room temperature, away from light and moisture. Do not store in the bathroom. Put the desiccant in the bottle. After opening the bottle, please use or discard within 3 months. Keep all medicines away from children and pets.
